For me just -DDEBUG at command line doesn't work. I do it by changing
/sys/modules/linux/Makefile (add -DDEBUG to CFLAFS). And then compile
and install the module:

#cd /sys/modules/linux
#make clean && make && make install

To use a new version you just do:

#kldunload linux.ko
#kldload linux.ko
