Ok, I used ifconfig_em0_ipv6="up" and it worked. So it looks like this:

ipv6_activate_all_interfaces="NO"
ipv6_network_interfaces="em0"
ifconfig_em0_ipv6="up"
ipv6_addrs_em0="2001:6a0:1cb::1-ff/64"
ipv6_defaultrouter="2001:6a0:1cb::ffff"
